About Enclomiphene

Mechanism of Action: Enclomiphene is a selective estrogen receptor modulator (SERM) that blocks estrogen feedback at the hypothalamus, increasing endogenous gonadotropin (LH and FSH) release and stimulating natural testosterone production.


Indications & Off-Label Uses: Investigated for male hypogonadism to increase testosterone while preserving fertility. Off-label, it is used as an alternative to exogenous testosterone for men seeking symptom improvement without suppressing spermatogenesis.


FDA Status: Not FDA-approved; clinical development has been ongoing.


Clinical Evidence: Studies show that enclomiphene can raise testosterone to physiological levels, improve libido, energy, and mood, and maintain sperm counts compared with exogenous testosterone therapy. Evidence is limited to small trials.


Safety & Monitoring: Generally well tolerated; reported adverse effects include headache, visual disturbances, and mild gastrointestinal symptoms. Monitoring of testosterone, LH/FSH, and hematocrit is recommended.
